RP?HPLC法测定钩藤煎剂中钩藤碱含量及变化

【摘要】 [目的]测定钩藤中钩藤碱在沸水后下煎药法中煎出量及变化规律。[方法]采用RP?HPLC法,Hypersil BDS C18柱,(150mm×4.6mm,5μm),以甲醇∶水(70∶30)为流动相,流速1.0ml/min,波长254nm,柱温25℃。[结果]钩藤碱在0.19μg~0.95μg范围内与峰面积线性关系良好r=0.9990。[结论]钩藤在沸水后下煎药法中,其钩藤碱在汤药中的含量在11.4min达高峰,其后汤药中钩藤碱的含量逐渐降低。

RP?HPLC Method Measure the Contents and The Regularity of Rhychophyline in Ramulus Uncariae Cum Uncis Liu Qinghua, Ge Erning Zhejiang University of Traditional Chinese Medicine, Hangzhou (310053), China

Abstract:[Objective]To measure the contents of rhychophyline in uncaria,and its regularity of variety with decoction time. [Methods]RP?HPLC method was chromatographic conditions were listed below: Hypersil?BDS C18 column (150mm×4.6mm.5μm), amobine phase composed of metranol?water(70∶30),a flow rate of 1ml /min and wave lengths was 254nm,column temperature was 25 ℃. [Results]In the range of 0.19μg ~0.95μg, the linear was very well between the content of rhychophyline and peak area. [Conclusion]The contents of rhychophyline in decoction reached the peak at 11.4min after uncaria was added in boiling water.

钩藤,性味甘凉。具有清热平肝,息风止痉的功效。临床主治头痛、眩晕、惊痫抽搐等症[1]。本文对钩藤采取沸水后下的煎药方法,分时采样,再用HPLC法分别测定在不同时间段钩藤碱在汤药中的`含量及变化规律。

1 仪器与试药

美国Waters高效液相色谱仪,484紫外检测器,510泵,浙江大学N2000色谱工作台,甲醇为色谱醇,水为双蒸水。钩藤碱对照品购于上海融禾医药科技有限公司(批号:071209),钩藤药材购于浙江中强医药有限公司。

2 色谱条件

色谱柱:Hypetsil BDS C18柱(150mm×4.6mm,5μm),流动相:甲醇∶水=70∶30,流速:1ml/min,检测波长:254nm,柱温:25℃。

3 方法与结果

3.1 对照品溶液制备 精密称取钩藤碱对照品4.75mg,置于10 ml量瓶中,用甲醇稀释至刻度,得对照品储备液。将对照品储备液按不同比例稀释,分别得质量浓度为47.5μg/ml、38.0μg/ml、28.5μg/ml、19.5μg/ml、9.5μg/ml的溶液,待用。下载论文

3.2 供试品溶液制备 称取钩藤药材15g 备用。取1只200 ml的烧杯加入100ml水,调节电压使电炉处于高温,武火至沸,迅速加入钩藤药材15g,开始计时,于1min开始取样,然后在30min内,每隔2min取1次样,每次2ml,上述实验重复3次,并对应混合3次所取样品溶液,共提取15种待测样品经离心,取上清液,再经0.45μm微孔滤膜过滤过,取续虑液,即得供试品液,备用。

3.3 线性关系考察 精密吸取上述各种对照品溶液各20μl,进样,按上述色谱条件测定,记录色谱图(见图1),以峰面积y为纵坐标,进样量x(μg)为横坐标,绘制标准曲线,并计算得回归方程为y=1130.1763x-53.1639,r=0.9990(n=5)。结果表明,钩藤碱进样量在0.19μg~0.95μg范围内与峰面积具有良好的线性关系。
